Determination of the Effect of 2,3-Dihydroxybenzoic Acid on Lipid Profile of Prostate Cancer Cells by Shotgun Lipidomic Analysis and Anticancer Activity

Abstract :

In this study, we aimed to determine the anticancer activities of 2,3-dihydroxybenzoic acid (2,3-DHBA) against PC-3 human prostate cancer cells. In addition, the effects of 2,3-DHBA on the lipid profile of PC-3 human prostate cancer cells were also investigated by the shotgun lipidomic approach. The anticancer activity of 2,3-DHBA which is a naturally occurring type of phenolic acid on PC-3 human prostate cancer cells was determined by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) method. In addition, the apoptotic effect of 2,3-DHBA was investigated by DAPI (4,6-diamidino-2 phenylindole) staining. Our results showed that 2,3-DHBA have an anticancer activity to PC-3 human prostate cancer cells and this effect was increased in dose and time-dependent. In addition, DAPI staining showed that apoptotic properties such as chromatin condensation and nuclear fragmentation increased in a time-dependent manner. Certain diseases, including cancer, are known to be associated with specific changes in particular lipid species. In the last step of our study, the effects of 2,3-DHBA on PC-3 human prostate cancer cell lipid profile were investigated using a shotgun lipidomic approach. As a result of the analyzes, it was observed that 2,3-DHBA application caused changes in fatty acids, glycerolipid, glycerophospholipid, sterol lipids, prenol lipids, and polyketide lipid species in PC-3 prostate cancer cell lipid species.
