Abstract :

In this study, it was aimed to investigate the effects of Consolida orientalis on Acanthamoeba castellanii (A. castellanii) cyst and trophozoites and cytotoxic potentials of plant extracts. Axenic cultures of A. castellanii trophozoites and cysts were prepared to test the amibicidal activity of Consolida orientalis. Consolida orientalis concentrations of 5 mg / 100 µL, 2.5 mg / 100 µL, 1.25 mg / 100 µL and 0.6 mg / 100 µL were prepared in 24-well plates and A. castellanii cysts and trophozoites were cultured in these mediums. Parasites were counted in cell counting device at 0, 1, 3, 6, 12, 24 and 48 hours after staining with trypan blue. In addition, the cytotoxicity of Consolida orientalis on the WI-38 human fibroblast cell line was tested with the XTT test. When A. castellanii, which was exposed to different concentrations of Consolida orientalis flower extracts, was examined, no live cyst and trophozoite were found at the concentrations of 5 mg / 100 µL, 2.5 mg / 100 µL after 24 hours. Aerial part of Consolida orientalis, after 12 hours, all trophozoites died at concentrations of 5 mg / 100 µL, 2.5 mg / 100 µL. And no viable parasitic cyst was found at concentrations of 5 mg / 100 µL at the end of 24 hours and 5 mg / 100 µL and 2.5 mg / 100 µL after 48 hours. As a result of the cytotoxicity studies, it was determined that Consolida orientalis did not show any cytotoxic effects on the WI-38 human fibroblast cell line, even at a concentration of 5 mg / 100 µL. Our data is thought to be a pioneering study to develop a new therapeutic agent in the treatment of diseases, the cause of which is A. castellanii.
